Oocytes are highly specialised cells, but the mechanism by which they acquire and retain their cell fate remains incompletely understood. Sarkar et al. now show that the gene repressive trimethylation of histone H3 lysine 9 (H3K9me3) contributes to oocyte formation in Drosophila melanogaster and identify a role for the nucleopore complex (NPC) in the maintenance of H3K9me3 and gene silencing in the egg chamber.
